E. colimutants that fail to grow at 20°C were isolated, and three such mutants were studied in detail. Two of these are defective in 50Sribosome biosynthesis at low temperatures and accumulate 43Sand 32Sparticles, respectively. The third mutant produces reduced amounts of 50Sribosomes and accumulates 21Sparticles at low temperatures. The 43Sand 32Sparticles contain 23SRNA and appear to be precursors of 50Sribosomes. The 21Sparticles contain 16SRNA. The mutation that causes accumulation of 32Sparticles is linked to thespclocus. Isolation and characterization of coldsensitive mutants should provide a fruitful approach to the study ofin vivoribosomal assembly.
