Substance P (SP) is synthesized and used by projection neurons of the neostriatum (NS) and is affected by neurological disorders such as Parkinson’s disease.’,’Previous studies have shown that afferent nigrostriatal dopamine (DA) neurotransmission exerts a positive control on striatal SP bio~ ynthesis.~.~ Because the NS also receives a substantial serotonergic innervation from midbrain raphe cell groups, we sought to determine whether the manipulation of serotonin (5-HT) neurotransmission would also alter the biosynthesis of striatal SP.In order to suppress 5-HT, adult rats receivedp-chlorophenylalanine [pCPA, an inhibitor of tryptophan hydroxylase (TPH)] for two weeks (100 mg/kg/daypCPA sc via an Alzet minipump) and were sacrificed at various times during and after drug treatment. DuringpCPA administration, steady-state levels of the mRNA coding for preprotachykinin (PPT, the prohormone precursor to SP) were decreased more than 80%(FIGURE 1) as determined by Northern blot analysis, but they recovered substantially (65% of control) one week after drug treatment ended (data not shown).
