The Na+-H+ antiporter is present in many different cells, and plays a central role in regulating intracellular pH. Activation of the antiporter occurs in response to a wide variety of growth factors and is thought to play an important role in the initiation of cell growth and division.'A wide variety of in vivo physiologic maneuvers (chronic acidosis, parathyroidectomy, glucocorticoids) stimulate the activity of the Na+-H+ antiporter in renal proximal tubule cells by increasing the V,,,. z Parathyroid hormone decreases the activity of the Na-H antiporter in a continuous cell line from the opossum kidney.'These previous studies have established that the regulation of the activity of the Na+-H+ antiporter is of major biologic importance. Recent studies have demonstrated modifier sites on the internal4 and external5 aspects of the renal Na+-H+ antiporter. It appears that the internal modifier site may play an important role in the acute regulation of Na+-H+ antiporter activity. 6 The purpose of the present studies was to investigate the effects of agents that may affect the Na+-H+ antiporter at the external modifier site.
