Streptococcus mutansis the primary cariogen that produces several virulence factors that are modulated by a competence-stimulating peptide (CSP) signaling system. In this study, we sought to determine if proteases produced by early dental plaque colonizers such asStreptococcus gordoniiinterfere with the subsequent colonization ofS. mutansBM71 on the existing streptococcal biofilms. We demonstrated thatS. mutansBM71 colonized much less efficientlyin vitroon streptococcal biofilms than onActinomyces naeslundiibiofilms. Several oral streptococci, relative toA. naeslundii, produced proteases that inactivated theS. mutansCSP. We further demonstrated that cell protein extracts fromS. gordonii, but not fromA. naeslundii, interfered withS. mutansBM71 colonization. In addition,S. mutansBM71 colonized more efficiently on thesgcprotease knockout mutant ofS. gordoniithan on the parent biofilms. In conclusion, proteases of early colonizers can interfere with subsequent colonization byS. mutansin vitro.
