GH and prolactin of rat anterior pituitary were fractionated by polyacrylamide disc electrophoresis. In vitro incorporation of 14C-leucine into these hormone fractions was measured in male and female rats at ages ranging from 14 to 120 days. Their rates of synthesis and release were found to change markedly during growth, and to be different between sexes. Generally, synthesis and release of GH were greater in males, whereas synthesis and release of prolactin were greater in females. The rates of synthesis and release of GH increased more rapidly in males, and those of prolactin more rapidly in females after 30 days of age. In male rats, GH synthesis was high for 7 weeks after birth, during which time the body weight increased rapidly; thereafter the rate decreased with age. There was no change in GH synthesis in female rats with age. Prolactin synthesis increased with age in both sexes, at a greater rate in females than in males.
