Tuber initiation in the potato plant (Solanum tuberosum L.) var Kennebec, appears to be promoted by a graft transmissible stimulus formed or activated by specific conditions of temperature and photoperiod. The stimulus proved to be persistent even after moving the plants to conditions unfavorable for tuber formation. Profuse tuber formation was achieved when the plants were kept at 20[degree]C during 8 hr. of daylight followed by a dark period at 14[degree]C for 16 hr. (inductive environment). No tubers formed when the plants were kept at 26[degree]C for 8 hr. of day-light, followed by 8 hr. of artificial light (1000f.-c.) at 20[degree]C, and 8 hr. of darkness at 20[degree]C (non-inductive environment). Leafy cuttings when planted in vermiculite developed tubers at the leaf axils, 7 to 14 days after planting if coming from induced plants, but no tubers formed if the cuttings were excised from plants growing under non-inducing environment. A leafy shoot from an induced potato plant used as donor, when graft united to a non-induced cutting as receptor caused the latter to tuberize seven days after planting the grafted cutting under non-inducing con-ditions. Stem pieces 2 cm. long with an axillary bud were dis-infected with mercuric chloride, 0.3% for 1 min. followed by 5 min. in 10% Chlorox, and grown in sterile culture. Stem pieces from induced plants formed tubers at the axillary bud when grown in 3% sucrose in 1% agar medium, and in 3% sucrose plus inorganic nutrients in 1% agar. No tubers formed in 1% agar. Stem pieces from non-induced plants developed shoots from the axillary bud when planted in 3%, 5%, 10% sucrose in 1% agar medium. No shoot growth took place when planted in 1% agar, and 20% sucrose in 1% agar medium.
