A novel water-soluble paracyclophane, CP44 (1), was designed and synthesized as a host molecule possessing a hydrophobic cavity to capture organic guests in water. X-ray crystallographic study revealed the formation of a 1:1 inclusion complex, and not a simple stacked complex, between protonated CP44 and durene. This is the first direct evidence showing the ability of a water-soluble cyclophane to form an inclusion complex. The inclusion of the hydrophobic guests in water (acidic condition) was also evidently observed by several kinds of spectra (1H and 13C NMR, fluorescence).
