Histamine was first observed to be a potent vasoactive substance by Dale and Laidlaw in 1911 (1) but was not associated with tissue mast cells until 1953 (2). Over the past 75 yr, histamine has been studied extensively, particularly in the past decade since the appreciation of histamine's actions through Hl and H-2 receptors has become clear. In this review, we will focus upon current concepts of the biosynthesis and biodegradation of histamine, the physiology of its release from mast cells and basophils, and the actions of histamine on target cells and tissues. In the course of this discussion, we will focus on the role of histamine in asthma and the mechanisms by which histamine may contribute to the physiologic alterations seen in asthmatic patients.
