MethodsIn our experiment we aimed to employ a method for the estimation of capillary permeability, which in itself does not alter the permeability inducing the liberation of histamine-like substances and at the same time it is suitable to analyse the time-conditions of the permeability increasing effect. Therefore we chose the method of Amster and Huber (1). According to this method the fluorescein-sodium in 0.5% solution (0.3 ml per kg body weight) was ad ministered intravenously. It penetrates through the blood-aqueous barrier and can be demonstrated with optical methods in the aqueous humour. As a quanti tative characteristic of capillary permeability we noted the duration of time during which the dye ‘saturated’the aqueous humour (the colour concentration of the aqueous humour reached an approximately maximal value). Thus, the shorter ‘saturation time’indicates an increased permeability. The experiments were performed on 120 albino rats of both sexes of our own breed, with a body weight of 180 to 250 g. The animals were anaesthetized with Nembutal (35 mg/kg body weight, ip). The endotoxin was extracted by the method of Westphal et al.(7) on various types of Escherichia coli strains.
