The reagent fluorescamine has been used for the fluorometric assay of proteins based on their content of free amino groups. A manual assay is reported which is applicable to 500 ng of protein. A semiautomated assay, which separates protein from low molecular-weight amines, has been used to assay 10–50 ng of protein. Applications of the reagent to the monitoring of proteins during enzyme purification are described.
