The radial distribution of electron density in filamentous bacterial viruses is calculated from the equatorial intensity on X-ray fiber diffraction patterns, at a resolution of 15 Å. The radial density distribution shows a cylindrical molecule with a central high density core 25 Å in diameter (interpreted as DNA), surrounded by a shell with inner diameter 40 Å and outer diameter 60 Å (interpreted as protein). Both class I and class II virions have similar radial density distributions. Other types of model are shown to be unlikely. In dry fibers, the periphery of the virion is distorted to enable the virion to pack in a hexagonal lattice with lattice constant a≅ 55 A ̊. The cylindrical model is substantiated by diffraction experiments on fibers treated with heavy atoms, and by electron microscopy of sections taken perpendicular to the fiber axis.
