Immunofluorescence microscopy allows one to detect microprojections on the lymphocyte surface and to follow their modulation by various environmental conditions. Both T and B cells express villi similarly. Among other factors, the expression of villi is decreased after cell chilling, is enhanced in the presence of metabolic inhibitor, is decreased in the presence of an agent affecting microfilament integrity, is relatively insensitive to agents affecting microtubule integrity, is sensitive to the pH, is inhibited by high does of lectins and is dependent on the lack of detectable crosslinking of membrane antigens by the ligand.
