Movements of the chloroplasts were studied in two marine centric diatoms: in Ditylum brightwellii the chloroplasts move outward and inward during the light-dark cycle, and in Odontella regia the chloroplasts are stationary in the cytoplasm. Physical relationships between chloroplasts and cytoskeleton were determined by immunofluorescence microscopy and electron microscopy, and the possible involvement of the cytoskeleton in the movements of chloroplasts was investigated using cytoskeleton-inhibitors. The results show that microtubules and microfilaments surround the chloroplasts of D. brightwellii and are involved in their movements. Neither microtubules nor microfilaments have a close relationship with the chloroplasts of O. regia.
