Retrograde reaction was studied in spinal motoneurons of Ranacatesbeiana. The lumbar segment of the spinal cord was removed 8-11 days after unilateral ventral root amputation (8th, 9th, 10th, sometimes 11th root), and sections were treated according to the following techniques Nissl thionin-blue stain, Bodian''s protargol impregnation, Gomori''s method for histochemical detection of acid phosphatase. Well-known cellular reactions following axonal section were confirmed in the motoneuron perikarya on the operated side: chromatolysis, decreased affinity for silver and neurofibrillar disintegration, heightened acid phosphatase activity. Similar alterations were traced further in the motoneuron dendrites. The characteristic pattern of dendritic processes in amphibian spinal cord, i.e. long expansions branching profusely through the white matter in the ventrolateral column and contributing to a dense marginal plexus under the pia, permitted the identification of retrograde effects extending to the entire length of the motoneuron dendrites. Dissolution of Nissl substance and neurofibrillar disintegration in the proximal shaft of the dendrites were found associated with chromatolysis in the parent cell-bodies. Decreased affinity for silver and heightened acid phosphatase activity were observed in the whole dendritic arborization, including the perimedullary plexus. Other experiments revealed an increased acid phosphatase reaction in the ventro-lateral column as early as 20 hours after axotomy. The results suggest an extensive participation of dendrites in various processes of the retrograde reaction.
