Gravid Fischet rats were given 100,000, 40,000, 25,000, or 10,000 IU/kg/day vitamin A palmitate (VA) orally on days 8–10 of gestation. The largest dosage produced high prenatal and postnatal mortality rates and marked growth retardation and abnormal gait in the few survivors. The three lower dosages produced no increased mortality, no weight reduction, nor any visible abnormalities in the offspring. The off spring were tested at 70–72 days of age in an open field. Those exposed to 100,000 IU VA showed hypoactivity compared with saline and nontreated controls. At 73–77 days of age the offspring were tested in a Y-maze avoidance task. All experimental groups showed decreased avoidance and discrimination acquisition, which was most pronounced in the 100,000 IU group.
