Publisher Summary Excitatory amino acid (EAA) receptors are currently divided into two major categories according to their signal transduction processes in the mammalian central nervous system (CNS), including the ionotropic and metabotropic classes. The metabotropic EAA receptor family so far comprises at least eight different subunits (mGluR1-8) and is subclassified into three groups based on differences in structure and signal transduction systems. On the other hand, ionotropic EAA receptors linked to cation channels are pharmacologically and physiologically classified into two major categories according to differential sensitivities to N-methyl-D-aspartic acid (NMDA). The NMDA channel is highly permeable to Ca2+, with different ion channels coupled to non-NMDA receptors. This chapter discusses a receptor binding assay, an electrophoretic mobility shift assay (EMSA) and an immunoblotting analysis for evaluation of signal transduction from membranes to nuclei.
