Ca45 disappeared from the medium when dog erythrocytes were suspended in buffered 0.16 [image] KC1 but not in 0.16 [image] NaCl. Two-day-old cells took up even greater amounts of Ca45 in KC1, whereas with aged cells in NaCl there was no detectible uptake. Thus, a difference in Ca45 uptake by dog cells was noted in the presence of K+ in contrast to Na+, as well as greater uptake by stored than by fresh cells in KC1. Uptake in KCl was greater at relatively alkaline pH. Acid-citrate-dextrose and, to a certain extent, heparin appeared to delay the increased uptake of Ca45 by aged cells. Addition of glucose, adenosine, or Nembutal [pentobarbitol sodium] to stored blood had no effect. Fresh cells partially hemolyzed by hypotonic media or by saponin took up no more Ca45 than unhemolyzed fresh cells. Cat and human cells failed to show significant Ca45 uptake. These results indicate an apparent competition between Na+ and Ca++ for binding sites in dog erythrocytes possibly as a consequence of the similarity in ionic radii.
