Two colorimetric procedures, the thiobarbituric acid and alkali-Ehrlich, have been developed for the determination of free sialic acid in the presence of bound sialic acid. These methods apparently involve reactions with different moieties of the molecule. Complementing each other and the existing methods of determining total sialic acid in sialo-mucoids, they serve as valuable tools in the study of the degradation and release of sialic acids from sialomucoids. The structural implications derived from the application of these tests are briefly discussed.
