Although spectrometric estimation of chlorophyll pigments using trichromatic equations (trichromatic method, STRJCKLAND & PARSONS 1972) has been known to be involved in considerable errors (eg MARliR et al. 1980), it has been widely done because of laek of the simple and reliable method. A shortcoming of this trichromatic method is that 90% acetone or 100% methanol extracts from phytoplankton samples contain various accessory pigments and degradation products of chlorophyll pigments which have the absorption spectra overlapping with those of chlorophyll pigments. Thus the best way would be to determine each chlorophyll pigment after chromatographic separation.In recent years with the development of chemically bonded stationary phase and high pressure pumps, high performance liquid chromatography (HPLC) has become one of the most efficient and reliable separation techniques, and is receiving a wide acceptance for the determination of chlorophyll pigments using reverse phase column (eg MANTOURA & LLEWELLYN 1983, BIDIGARE et al. 1985).
