‘I ‘remendous idwinces have been made w ithin the last decade in our knowledge of the cellular and molecular endocrinology of hormone-responsive carcinogenesis. pilrticularly breast cancer. Hoth homiones and 1iormone-indirc. ible autocrine growth factors play a prominent role in the mechanisms rnediating the growth of this important neoplasm of the reproductive system. In fjict most breast cancers probably rely to some degree upon oestrogen, oestrogen-inducible growth factors and/or prolactin for their growth [1, 21. hlelatonin, the m; i. ior secretory product of the rnammalian pineal gland, is an important neuroendocrine regulator of reproductive physiology in many mammalian species. In R number of these species including the rat, this indoleamine inhibits the development and growth of the normal reproductive tract 13].‘I’herefore. me hiive been using animals with carcinogen-indiIc. td mammary cancers in viva, as\\ell; is hum; in breast cancer cell lines in 7ritr0, as model systems for studying the neuroendocrine role of melatonin in oncogenesis 1-1. The following discussion mill review some recent findings that support the hypothesis that melatonin is an oncostatic hornioiw by virtue of its iibility to inhibit the mitogenic action of steroids;~ nd polypeptide hormones and growth tictors.
