When developing Dictyostelium cultures are disaggregated, the amoebas are capable of recapitulating the morphogenetic stages they had previously progressed through in approximately 1 10 th the original time. When developing cultures are disaggregated and placed in growth medium, they lose this capacity for rapid recapitulation in a discrete step which we previously called “erasure.” In this report, several characteristics of the erasure phenomenon are analyzed, and data are presented which demonstrate that:(1) the timing and nutrient requirements of erasure change as cells progress through development; and (2) the capacity for the rapid recapitulation of different stages can be erased at different times, indicating that events which influence the recapitulation timing of different stages are dissociable. For cells derived specifically from the tight aggregate stage of morphogenesis, we have presented data which indicate that:(1) erasure depends upon a period of de novo protein synthesis which begins 30 min after cells are inoculated into nutrient medium and which extends to the moment of erasure;(2) a period of stabilization for the erasure event (s) occurs within 50 min after erasure and depends upon de novo protein synthesis;(3) inhibitors of transcription do not significantly affect the erasure time of later aggregation stages, but retard the erasure of an earlier stage; and (4) erasure occurs approximately 2 hr after the reinitiation of cell growth and several hours prior to the first synchronous cell division in complete nutrient medium. The complexity and possible nature of both the capacity to recapitulate rapidly and the erasure phenomenon are discussed in light of these observations.
