T cells expressing either the al3 or yS antigen receptor (TCR) are distinct cell lineages. The single locus encoding the TCR a and 6 genes requires special regulation to avoid a gene expression in y6 T ceils. We show here that the minimal a enhancer is active in the y6 T cell lineage but gains af3 lineage specificity through negative &-acting elements 3’of the Ca gene that silence the enhancer in $3 T cells. The negative elements at the Ca locus consist of several silencers that work in an orientation- and distance-independent fashion. These silencers also act on a retmviral enhancer that is normally ubiquitously expressed, restricting its activity to afl cells. The a silencers are active in non-T cell lines, suggesting that the decision of a cell to differentiate into the aj3 T cell lineage may involve specific relief from these silencers. Silencers are likely to be as important as enhancers in establishing lineage-specific gene expression in many systems.
