T-cell receptor (TCR) complex is a heterodimer composed of an α-and a β-chain (TCRA/TCRB complex) or a γ-and a δ-chain (TCRG/TCRD complex). The genomic organization of TCRA, TCRB, TCRG and TCRD genes and their chromosomal localization have been determined in human and mouse (O'Brien 1993). In artiodactyls, an order representing the most dramatic radiation of large mammals, the TCRG/TCRD complex represents the predominant form of the peripheral TCR pool (Hein et al. 1993). However, little information is available on the genomic organization and chromosomal localization of these genes in these species. Recently, in sheep, the TCRA/TCRD locus has been assigned to chromosome bands 7q1. 4±q2. 2 (Massari et al. 1997). Moreover, analysis of γδ TCR cDNAs indicates that the sheep genome contains® ve functional Cγ segments (Cγ1±Cγ5)(Hein et al. 1993). In the present paper, we report isolation, molecular characterization and cytological mapping of sheep genomic clones containing Cγ1 and Cγ2 constant genes.
