AMANTADINE HYDROCHLORIDE was first reported to be effective in the treatment of Parkinson’s disease by Schwab et a1.l in an uncontrolled trial done in 1969. In subsequent controlled studies, Parkes et al.2 reported that of 35 patients treated, most showed clinical improvement. Using higher doses in these patients, an additional response was ~ b t a i n e d . ~ On the other hand, Hunter et al.4 found that amantadine had no significant effect on functional disability. In all of the above studies, amantadine was given in combination with the patient’s previous antiparkinsonian medications. In the present investigation, the effectiveness of amantadine hydrochloride ( Symmetrela) alone was established in a doubleblind crossover study, and its effects, upon routine laboratory tests, electroencephalography (EEG), and amine metabolism were determined.
