The formation of the active amino acyl-RNA-messenger RNA-ribosome complex is an enzymatic reaction. The "binding" enzyme and GTP are required. The movement of the messenger RNA in relation to the ribosome requires peptide bond synthesis. Since uncharged transfer RNA does not participate in the binding reaction, after a peptide bond is formed, presumably the uncharged RNA leaves the ribosome by diffusion rather than by an exchange mechanism. This mechanism differs considerably from that proposed for E. coli. The major experimental differences are the nonenzymatic nature of the binding and the participation of uncharged transfer RNA.
