Adalimumab (Humira®), an anti-TNF-α monoclonal antibody, leads to highly variable serum concentrations between patients [1, 2]. Although an important portion of this variability may be due to the subcutaneous route of administration, the variability of adalimumab absorption kinetics following a subcutaneous injection has never been reported. In the Novel Therapies for Resistant FSGS (FONT) phase I study [3, 4], patients with focal segmental glomerulosclerosis (FSGS) were randomized into two arms that evaluated adalimumab [4] or rosiglitazone [3]. We report a description of adalimumab PK in FSGS patients using population compartmental modelling, with a focus on evaluation of adalimumab absorption kinetics.
