Outline of diacylglycerol metabolism in animal cells. Depending on physiological conditions of cells, DG can potentially be generated from five sources:(1) from PA in de novo synthetic pathway;(2) from phosphatidylinositols (phospholipase C);(3) from PC (phospholipase C);(4) from TG (lipase); and (5) from 2-MG (acyltransferase). The role of phospholipase C acting on PIP 2 has been well established, but the contribution of other pathways in generating DG remains to be defined. Upon ceil stimulation DG is phosphorylated by DGK (6) to yield PA, which is then used for resynthesis of phosphatidylinositols via CDP-diacylglycerol (7). DG may be hydrolysed by a lipase to 2-MG (8), which is a substrate in vitro of DGK (9). For simplicity details of reactions and those not directly relevant to the present discussion are omitted.
