By means of synaptic stimula-tion or by applying a depolarizing current through an intracellular electrode, motoneurones of anesthetized cats were caused to discharge impulses which were recorded in ventral root filaments. This dis-charge was always associated with a spike in the initial segment of the motoneurone (the IS spike), but was quite independent of the somadendrite spike (the SD spike). Also, the impulse appeared in the ventral root so quickly after the onset of the IS spike that it must have been initiated either by the earliest portion of the IS spike or even just before by a primary response of the 1st node of the medullated axon. When allowance is made for accommodation, it appears that depolarizations produced by synaptic action or directly by applied currents are equipotent in generating motoneuronal spikes. The threshold depolarization for the initial segment varied from 5-18 mv, with a mean of 10 mv, for moto-neurones relatively undamaged by the micro-electrode, while for the soma-dendritic membrane the range was 20-37 mv. The relationship of these findings to the functioning of the motoneurone is discussed.
