In the past few years, there has been increasing evidence that much of molecular evolution may not involve neutral alleles. This is almost certainly the case from protein5 and may be true for silent sites in coding regions as well.9, 14 Should we feel compelled to reject the neutral theory for certain classes of molecular variation, there is a conspicuous vacuum in the literature when we seek alternative models. The time seems ripe, therefore, to begin an exploration of non-neutral models to see how compatible they are with the very observations that cast doubt on the neutral theory.
