There are three issues of particular interest in studies of DNA polymerases. First, what role does the enzyme play in assuring the fidelity of template-directed DNA synthesis? How does the enzyme reduce the incorporation of incorrect bases and detect and remove mismatch base-pairs? Second, what accounts for the enzyme's processivity, ie the successive incorporation of nucleotides without enzyme dissociation? Third, how might we imagine that prebiotic polymerases evolved? Escherichia coli DNA polymerase I was first isolated by Kornberg and colleagues and has become a most suitable enzyme for understanding how DNA polymerases work (Kornberg, 1980). Pol1 is a 103 000 dalton monomer, has DNA polymerase, 3'-5'exonuclease and 5'-3'exonuclease activity and has separate binding sites for deoxynucleoside monophosphate and triphosphate
