SUMMARY Humanmacrophage agglutination factor (MAggF)isalymphokine witha numberofbiochemical andimmunochemical similarities tothefibronectins (FN). Thepresent study was undertaken to define this relationship more exactly. Peripheral bloodmononuclear cells (PBMC)andclonedhuman CD4+T-cell lines synthesized bothMAggFandimmunoreactive FN denovo following antigen or mitogen activation. MAggFproduced underthose conditions co-purified withan immunoreactive FN on gelatin-affinity andgel-filtration chromatography. ThisFN hadaconsistently slightly smaller relative molecular mass thanplasma FN on bothimmunoblotting andgelfiltration. Purified human MAggFwas extremely active: itagglutinated humanmonocytes atfM concentrations andwas upto 2,000,000 times more active inagglutinating mononuclear phagocytes thanotherpurified FN.The action ofMAggFon monocytes was dependent on atleast twoantigenically distinct cell-surface FN receptors. We suggest thattheextremeactivity ofMAggFisa result ofco-operative interactions between FN domains on theMAggFmolecules andmultiple distinct classes ofFN receptors on responding cells.
