Reviews the literature on the biochemical changes and pharmacologic effects during the opiate abstinence syndrome, focusing on areas where overlap is apparent between different biochemical mechanisms of withdrawal phenomena. These areas may provide clues to possible" final common pathways." The evidence for alterations of endogenous opiate receptors during opiate dependence, the involvement of some intracellular modulators, the evidence for alterations in 6 neurotransmitters, and some behavioral and environmental factors in opioid withdrawal that may have implications for cellular mechanisms are discussed. Considerable portions of the evidence, from endogenous opioid distributions and activities to the antiwithdrawal effects of clonidine in human Ss, suggest that central norepinephrine systems, including the nucleus locus coeruleus, may be important for further studies of the biochemistry of the classical opiate withdrawal syndrome. Involvement of several known intracellular, receptor, and neurotransmitter mechanisms in opiate withdrawal may be traced to interactions with noradrenergic neurons.(PsycInfo Database Record (c) 2020 APA, all rights reserved)
