Using various fractionation procedures the following distribution of magnesium in rat liver mitochondria was found: outer membranes, 4%. intermembrane compartment, 50%; inner membranes, 5%; matrix, 41%. The highest Mg/protein ratio was found in the intermembrane compartment. Incubation of mitochondria in the presence of ADP + 2,4-dinitrophenol releases most of the intermembrane magnesium and has little effect on magnesium content in other compartments. This process is atractyloside insensitive.
