METHODSRings of the aorta of the turtle and of the stomach muscle of the frog, both 2-3 mm wide, and longitudinal strips of the rat’s uterus were used. They were suspended in 10 ml solution. Tension was recorded on a polygraph using a Grass FT03 as transducer. The tension produced during a spontaneous cont. raction of preparations of the aorta was usually l-2 g.Ringer solution for cold-blooded animals contained (in mM): NaCl, 115; KCl, 2; CaC12, 1: MgSO4, 0.6; NaHCOs, 4; NaH2POa, 0.5. In experiments on the effect of pH only phosphate buffers were used. Mam-malian Ringer solution contained (in mM): NaCl, 140; KCl, 2.5; NaHCOz, 9; NaH2P04, 0.5, glucose, 5. Oxygen bubbled through the solution continuously. Ethylenedioxybis (ethyleneamino) tetraaceticacid (EGTA), obtained from Geigy Chemical Corporation, was added as 0.1 M solution adjusted to pH 7 by KOH. Ca-free solutions were changed twice at the beginning of an experiment at intervals of about 10 min. The concentration of drugs is given as gram base per milliliter. The experiments on aorta and stomach muscle were carried out at room temperature(24-26 C), those on uterine muscle at 35 C.
