This article describes a set of vector plasmids that are suitable for C-terminalin situepitope tagging ofTrypanosoma bruceigenes either by a single-step polymerase chain reaction (PCR) procedure or by conventional cloning via PCR-amplified fragments. This set of plasmids allows multiple protein tagging within a single cell. The set is expected to be useful for the intracellular localization of, and protein and protein-complex isolation inT. brucei.
