Ulcerative colitis (UC) is an inflammatory bowel disease of unknown cause and is characterized by cycles of acute inflammation, ulceration and bleeding of the colonic mucosa. Oxidative stress has been implicated to be a key factor in the pathogenesis and perpetuation of mucosal damage in UC (1). Glutathione (GSH) is a ubiquitous tripeptide-thiol present in high concentrations in mammalian cells, and is a significant component of the antioxidant defenses of most tissues. GSH exists either as reduced glutathione or oxidized glutathione (GSSG), and the cellular redox status is maintained by the distribution of GSH and GSSG predominantly as GSH. This redox balance is achieved by the function of the redox cycle enzymes. Exposure of cells to oxidant stress results in GSH/GSSG imbalance and the accumulation of GSSG. This study was undertaken: 1) to investigate the colon mucosal redox content and oxidation state of GSH in active and inactive UC states and 2) to examine the relationship between GSH status and efficacy of therapy in this patient population.
