A method is described for the dissection of the rat brain into the following regions: cerebellum, medulla oblongata, hypothalamus, striatum, midbrain, hippocampus, and cortex. The endogenous norepinephrine content of each region was measured; highest concentrations were found in the hypothalamus and lowest in the cerebellum. The distribution of radioactive catecholamines in the 7 regions was examined at different times after the injection of small amounts of tritiated norepinephrine, di-hydroxyphenylalanine (DOPA) or dihydroxyphenylethylamine (dopamine) into the lateral ventricle of the brain. After the administration of [H3] norepinephrine, the accumulation of the labelled amine in various brain regions correlated with the regional distribution of endogenous norepinephrine, highest levels being found in the hypothalamus. High levels of [H3]noreplnephrine were, however, also found in the striatum[long dash]a region which contained a high concentration of endogenous dopamine but only a low concentration of norepinephrine. There were no qualitative differences in the metabolism of labelled norepinephrine in the different brain regions, though quantitative differences in metabolites occurred. In most regions the proportion of metabolites to unchanged norepinephrine decreased with time except in cerebellum and cortex. Unchanged [H3]norepinephrine disappeared at different rates from the various brain areas. After the administration of [H3]dopamine there was a rapid formation of [H3]-norepinephrine inmost brain regions, but maximum levels of labelled norepinephrine were attained at different times. In the striatum only very small amounts of labelled norepinephrine could be detected, but the highest levels of [H3]dopamine were found in this region. The regional distribution of tritiated catecholamines after dopamine was similar to that found after [H3]norepinephrine. After the injection of [H3] DOPA there was a limited conversion to catecholamines; in all areas of the brain the levels of [H3]dopamine greatly exceeded those of [H3]noreptnephrine 5 hr after the injection of DOPA.
