MATERIALS AND METHODSFour children, ages 3, 5, 6, and 11 months, and 1 at 3 weeks of age, with clinically proved cystic fibrosis of the pancreas were surgically explored, in an effort to seek anatomic obstruction in pancreatic ducts and extrahepatic biliary tree. The operative procedures used were reported elsewhere. 20 At exploration, tissue was removed from the pancreas and liver for biopsies, and the gallbladder was removed.For light microscopy, portions of the biopsy specimens were fixed in 3: 1 acetic acid-methyl alcohol, embedded in paraffin, and sections stained by means of hematoxylin-eosin, mucicarmine, Pearl's iron stain, periodic acid-Schiff, Gomori, and Verhoff-Van Gieson stain technics. For electron microscopy small sections of tissue from pancreas and liver were immediately fixed in Dalton's osmic tetroxide solution7 for 1 hr. at 4 C. Embedding was performed either in a prepolymerized mixture of 8: 1 butyl-methyl methacrylate with the addition of benzoyl peroxide as a catalyst, or in Epon 812. 13 Sections were cut with a Porter-Blum microtome, using glass knives, and examined with an RCA model EMU-3F electron microscope. Some of the ultrathin sections were stained with lead hydroxide, uranyl acetate, and phosphomolybdic acid. 21 The gallbladders were examined only with light microscopy. As control for light microscopic studies of the fresh tissues, autopsy material from 7 patients with cystic fibrosis of the pancreas and 15 children with other diseases, but normal livers and pancreases, were reviewed.
