Sea urchin sperm flagella, after isolation and glycerination, are nonmotile, but dephosphorylate ATP at a rate which is 1 5 to 1 2 of the probable rate of ATP dephosphorylation in actively motile spermatozoa. Addition of O. 3 m thiourea to the ATPase assay solutions, extraction with O. 6 m KCl, and several other treatments which inhibit the motility of glycerinated spermatozoa, increase the rate of ATP dephosphorylation by 100% or more. This apparently represents an uncoupling of ATP dephosphorylation from other components of the system which generates movement. The uncoupled ATPase has a higher K m, greater specificity for Mg 2+ ion activation, and other altered characteristics in addition to increased activity. Although uncoupling and extraction usually occur together, under some conditions the ATPase activity can be extracted in a less modified form, which is still activated by thiourea. Further work is required to gain complete control over the uncoupling process and understand its implications.
