In separated canine renal tubules intra-cellular pH was calculated from the distribution of DMO [5, 5-dimethyl-2, 4-oxazolidinedione] 14C, using inulin as a measurement of noncellular water. At normal P(X>2 [carbon dioxide pressure] and extracellular [HCO3-] renal cells were found to be more alkaline (mean pH 7. 32) than most other mammalian cells. Cell acidity varied in direct linear relation to extracellular acidity when the latter was changed by systematic variation of either PCO2 or extracellular [HCO3-], but PCO2 had a significantly greater effect. Inhibition of carbonic anhydrase by addition of 10-5 M acetazolamide significantly increased cellular pH. Neither acute K depletion nor rapid repletion significantly affected cell pH. The results suggest that renal tubular cells are relatively alkaline because they secrete acid; the secretory process appears to be only indirectly linked to carbonic anhydrase. [long dash]Authors.
