MATERIALS AND METHODSThe tumor being investigated arose spontaneously as a renal adenocarcinoma in an inbred Balb C/Cr mouse. The study was begun when the tumor was in its sixth transfer generation. The tumor is transferred by a single cell suspension. The suspension is made by grinding 1 g of trimrned diced tumor in a sterile glass tissue grinder with 9 ml of sterile chilled 4 C McCoy’s medium (5% fetal bovine sera, penicillin, 100 units/ml, and streptomycin 50 pg/ml). The solution is passed through a 27-gauge needle and viability is determined by erythrocin staining[9]. The viability ranges consistently from 90 to> 95%. The concentration is then adjusted to 1 X 10” live tumor cells per ml. The animals receive a total standard dose of. l ml or 1 X 10” live tumor cells in one or two aliquots unless otherwise indicated. A 27-guagc needle and sterile I, $ cc cyringe are used for all tumor inoculations. The suspension is kept at 4 C and agitated frequently to prevent settling out of cells.
