PIII [a slow, large, negative potential change] response to light stimulus in the vertebrate was studied using Ba. K decrease near the photoreceptor could cause Mueller cells [MC] to hyperpolarize and elicit a PIII response. Ba decreases K conductance providing a test of the MC possible PIII response. Dark adapted bullfrog retina were separated from the pigment epithelium and perfused with Ringers solution buffered with Tris malate. Extracellular recordings of response to light stimulus [250 ms, 47 .mu.W/cm2] were made by ClAg electrodes embedded in the perfusion chamber under normal buffer conditions and in Ba Ringers solution [0.4 mM BaCl2]. The Ba solution suppressed the slow PIII response. After return to the normal solution the PIII response returned slowly [115 min partial return] contrasting to the photoreceptors rod and cone responses that returned fully within 20-30 min. The slow PIII response apparently occurs in the MC.
