Alzheimer’s disease is expressed in a three-fold nosology: anatomic, biochemical, and behavioral. Numerous reports have found similarities and interrelationships between Alzheimer’s disease and Down syndrome on all three levels, and epidemiological associations as well. It seems logical to ask whether the demonstrable relationship between Down syndrome and Alzheimer’s disease could reflect any similarity in mechanisms; that is, whether testable insights can be gained for both the etiology and pathophysiology of Alzheimer’s disease by considering the hypothesis that the disease shares a similarity of causality with Down syndrome Given that Down syndrome is a disorder whose genetic causality is well characterized, the question is whether Alzheimer’s disease may involve any genetic similarities.
