The effects of puromycin, high concentrations of thymidine, and actinomycin D on protein, deoxyribose, and ribose nucleic acid synthesis, respectively, were examined in connection with the repair of sublethal X-ray damage measured by the dose fractionation technique. Within limits set mainly by drug toxicity, it was found that neither puromycin nor thymidine affect repair, although actinomycin D at very low concentration (5× 10-9 M) does. The degree of inhibition of synthesis produced by these drugs were: puromycin, ∼ 60%; thymidine, close to 100%; and actinomycin D, 50%. However, the concentration of puromycin used (6.0 μg/ml) was found to inhibit RNA synthesis as effectively as a level of actinomycin D which had a pronounced effect on repair. Analyzed together with earlier results which showed that repair proceeds quite well at reduced temperatures and does not require oxygen, we propose that repair may be a passive (nonenzymatic) process involving deoxyribose nucleic acid in stereochemical relationship...
