We have cloned and sequenced a highly repeated satellite-type DNA sequence from the domestic pig (Sus scrofa). DNA bearing this sequence is visible in EcoR1 digests of genomic DNA as a band of approximately 1 kb. This band was isolated and cloned into pUC9. A clone now designated pM/F-1258 was sequenced using M13-dideoxy methods (Figure 1). The 1258 bp sequence shows imperfect intemal repetitions of about 44 bases. Sequences hybridizing to the cloned segment are equally well-represented in the genomic DNA of male and female pigs. We have use the cloned sequence as a gender-neutral probe in determining the sex of embryonic tissue samples (Figure 2).
