USE OF ACID, ROSE BENGAL, AND STREPTOMYCIN IN THE PLATE METHOD FOR ESTIMATING SOIL FUNGI

A study was made of the comparative use, singly and in various combinations, of acid, rose bengal, and streptomycin as bacterial growth inhibitors in various fungus media for estimating numbers and kinds of soil fungi by the dilution plate method. Concns. of 1:5000 to 1:30,000 of rose bengal prevented growth of most bacteria in some soils, while others contained sufficiently large numbers capable of developing at these concns. that fungus counts were decreased. A combination of rose bengal (1:15,000 or 1:30,000) and 30 ug./ml. of streptomycin virtually eliminated bacterial growth from all soils tested. Streptomycin alone slightly inhibited growth of 2 fungus cultures out of 117 tested. With few exceptions rose bengal had little effect other than reducing the size of the colonies. 17 fungi, which developed on media containing rose bengal alone or in combination with streptomycin, failed to grow or produced extremely poor growth on acidified agar (pH4). Peptone-dextrose agar containing 1:30,000 rose bengal and 30 ug./ml. of streptomycin was rated the best medium for soil-fungus plating. By use of this medium fungus counts were increased as much as 100% and the kinds of fungi isolated by 14% compared with the same medium acidified to pH4. Six fungi that were consistently isolated in relatively large concns. on the new medium were rarely or never found on the acid agar.
