Methods The gel chamber microtechnique developed by the author. The matrix is prepared by drilling holes of 3 mm. in diameter but with an aperture at the lower surface of 2 mm. making a suitable pattern in a piece of plexiglass (35x40 mm.) of a thickness (2-4 mm.) which will provide the desired basin volume. Strips of waterproof tape not more than 4 mm. wide are applied to the lower surface (on two opposite sides) of the matrix to provide a chamber of the desired depth (0.4-0.8 mm.).(If handled with moderate care such a matrix may be used many times.) Cleansed glass slides (50x50x 1 mm.) arc agar filmed by spreading about 0.2 ml. of 0.3% agar over each and permitting it to dry. The gel diffusion chamber (see fig. 1) is formed by placing a matrix on an agar filmed slide and allowing a sufficient quantity of 1% agar in 0.85% NaCl at 50-60 C to flow into the chamber from one of the open sides.
