The losses of K from the erythrocytes of several species of mammals in hypotonic solutions at 40[degree] were studied. All the species studied lose K under these conditions and their relative rates are in the order: rabbit, horse, ox > guinea pig> pig, human. The plot of rate of loss of K against the tonicity of the medium is S-shaped indicating that there is a critical volume which a given cell must attain before it becomes permeable to K. When the swollen cell loses sufficient K to allow of its shrinking to this critical volume the loss of K ceases from this cell, so that the final concn. of K in the cells when the loss of K has ceased is still much higher than that in the suspension medium. Subsequent dilution of the suspension medium causes the loss of K to start again. The effect is not peculiar to the temp. of 40[degree] but varies continuously with the temp., and in the case of the rabbit detectable losses occur at 20[degree]; an approximate estimate of the temp. coeff. in the case of this species gives a value of about 2. The possibility that the losses of K are due to heavy metal impurities in the saline solutions is excluded by direct expts.; it is concluded that they are due to a mechanical stretching of the erythrocyte membrane.
