Isolation and characterization of microsatellite DNA loci in Japanese flounder Paralichthys olivaceus (Pleuronectiformes, Pleuronectoidei, Paralichthyidae)

A (CA)n-enriched Japanese flounder genomic library was produced. 80 clones from this library were sequenced; 59 clones contained repeat sequences. PCR primers were designed to amplify 27 of these loci. Polymorphisms of 16 loci (Po1, Po13, Po20, Po25A, Po26, Po31, Po33, Po35, Po42, Po48, Po52, Po56, Po58, Po83, Po89andPo91) in 64-69 fish were analysed in detail. The number of alleles ranged from 4 to 40, the observed heterozygosity ranged from 0.43 to 0.99 and the expected heterozygosity ranged from 0.43 to 0.97. OnlyPo31displayed significant deviations from Hardy-Weinberg equilibrium. The nucleotide sequences reported have been deposited with the DDBJ/EMBL/GenBank databases with the accession numbers AB046745-AB046746 and AB046748-AB046761.
