The study of cellular and tissue function requires the ability to assay the molecular phenotype of cells under normal and pathophysiologic conditions. In the lung, very little is known about gene expression by normal cells and how they are changed in disease processes because of the difficulty in obtaining more than a small number of homogeneous cells and their failure to proliferate in culture. Experimentally, it is preferable to assay freshly isolated cells and tissues to avoid organ or cell culture artifacts. Clinically, the assay of small amounts of tissues or cells allows the temporal analysis of cellular physiology. In this Update, we discuss a new molecular technique for phenotyping the gene expression in a single cell or small numbers of cells. This technique has important applications for the study of lung biology. It should be
